Job Description
A PDRA post in protein crystallography is available from 1st June 2016 for a period of one year in the first instance, renewable for a further three years to work with Dr. Ehmke Pohl on an EU-funded project on the biophysical and structural characterisation of novel viral proteins. The Durham group is part of a multinational team led by the Icelandic company Prokaria with the goal to identify and characterise novel viral proteins isolated from a wide range of bacteriophages that thrive in extreme conditions.

Responsibilities
Primary technical responsibilities include the protein expression, purification and the biochemical, biophysical and structural characterisation.
Prepare work plan, keep comprehensive, accessible, secure records of experimental work, data and analyses.
Assist PhD students and group members in the delivery of project objectives and assist PhD students in problem solving.
Prepare quarterly reports; participate in scheduled face-to-face meetings including presentation of research results.
Organise synchrotron trips, collect diffraction data on site as well as by remote access.
Give research presentations to national and global conferences, contribute to the consortium meetings.
Write or assist with the preparation of papers for publication in global journals and conference proceedings.
Liaise effectively with all members of the research team, and the technical and research support staff.
The successful candidate will work closely with the technician who will support protein production and they will assist training/supervision of PhD students. The post-holder will report to Dr. Pohl as project lead and will liaise closely with the European partners.
Please note the position is fixed term for one year starting as soon as possible, and renewable for up to a further three years.

Desired skills and experience

Requirements
The post-holder will have specific responsibility for the following areas:
(a) Expression of novel viral proteins using recombinant production systems.
(b) Purification, biochemical and biophysical characterisation of the target proteins using a range of techniques including surface plasmon resonance fluorescence anisotropy, isothermal calorimetry and small-angle X-ray scattering
(c) Crystallisation and crystal structure determination of target proteins alone, and in complex with ligands including nucleic acids.
Person Specification
Essential
Expertise in recombinant protein expression, purification using modern chromatography methods and crystallisation.
Expertise in X-ray diffraction data collection using in-house sources and 3rd generation synchrotrons
The ability to solve and refine protein crystal structure using molecular replacement or SAD/MAD or MIR methods
Proven ability to conduct research at a world-leading level.
Excellent team-working and interpersonal skills.
A motivated, enthusiastic and organised self-starter; someone who can work with a minimum of supervision, yet at the same time contribute to an excellent research environment
Desirable
Experience in biochemical and biophysical characterisation of protein-ligand interactions.
Experience in biological small-angle X-ray scattering, from sample preparation to data collection, analysis and interpretation.
Experience in the preparation, biochemical analysis and crystal structure determination of protein-DNA/RNA complexes.
Additional expertise related computational fields, such biomolecular modelling, ab inito structure solutions, chemical pipeline software and molecular docking.
Skills in LINUX system management, and core skills of programming in Python or a higher programming language.
